The Wombats delivered their fans the memorable 33-point win with a captivating last-quarter performance.
Up by nine points at the last break, Lancaster stormed home to kick seven of the last 10 goals of the match.
Speedster James Ross booted two goals in the final term, while best-on performer Zac Cerrone registered a brilliant four-quarter display.
The 17.10 (112) to 11.13 (79) triumph leaves Lancaster unbeaten and atop the Kyabram District League ladder after seven rounds.
Lancaster coach Corey Carver hopes the win over the reigning premier instils confidence in his playing group.
“Hopefully it gives the group belief,” Carver said.
“We have had a relatively young group in recent years and a win like this will only benefit them going forward.
“Hopefully it shows them that they have what it takes against the best sides.
“We haven't beaten them in a while, I'm not sure on the exact dates, but it's been several years.
“They have always had the measure on us and it's nice to get on the winning end of one.”
The top-four encounter was billed as a contest that would deliver a thrilling match.
The game lived up to the hype with only a couple of kicks separating the two sides in the first half.
Billy Ryan snagged all five of his goals in the opening half and was again an important contributor to his side.
Nagambie — led by star mid-season recruit Jamason Daniels (three goals), who last played locally at Tatura in 2019 — held the upper hand against the home side and led by six points at half-time.
Inaccuracy hurt the Lakers in the third term, with five behinds all that the powerhouse side could muster as the Wombats hit the lead.
The home side could smell victory, its first against Nagambie since 2013, and sealed the win in classy style.
“Nagambie missed a few shots on goal at the start of the third and we were able to go forward and kick a couple,” Carver said.
“We could have very easily been behind at the last break. We did a big pre-season and you kind of hope that your fitness stands up in those type of matches.
“Zac (Cerrone) was consistent all day and Jimmy (Ross) was slow to start, but worked his way into the game and finished strongly.”
Tom Madden and Mark Nolan flew the flag for Nagambie and were the two standout performers.
Lancaster's win sets up another mouth-watering clash this weekend, with the Wombats set to face Avenel in a top-of-the-table bout.
