Stanhope will step out on the court in round two for the first time since the memorable 2019 grand final triumph over Dookie United.
The Lions sat out last week due to the bye and will host Undera at their home deck.
The round two contest will be playing coach Jamieson Young's first game at the helm after taking over from premiership coach Kerrie Gray.
Young, who missed out on the 2019 season due to pregnancy, said she and her young squad were eager to hit the court on Saturday.
“Everyone is really excited to get out and play. We were a bit jealous last week watching everyone play,” she said with a laugh.
“I'm pretty eager to check out how our squad goes. We have got a new-look, young side which is really exciting.
“And I can't wait to play, I missed 2019, so it's been a long time coming.”
But the team that lines up on Saturday will be entirely different to the one that clinched premier and champion status only two years ago.
Only premiership captain Molly Breen remains, while two juniors Jemma Wright and Olivia Quattrocchi were involved as development players.
Multiple Warren medallist Chicky Barr has departed, but the gaping hole left by the star shooter allows for a young talent to shine.
Which is what Young expects from players such as Wright and Quattrocchi.
“We only have one player from the starting seven in that grand final,” Young said.
“But our two development players from that squad, Jemma and Olivia, will take on a bigger role this year which is exciting for them.
“They have had that taste of A-grade netball, they got court time in 2019 and they'll be ready to go this year.”
Undera began its season with a tough 47-41 loss to Rushworth.

The Redskirts were dominant winners over Merrigum in round one, with Fiona Down's squad clinching a 20-goal victory.
Dookie United playing coach Maddison Owen, fresh off her side's stirring win over Shepparton East, said her side would work on its combinations and stick to its game plan on Saturday.
“Tally are always a tough match-up,” Owen said.
“We will stick to our game plan and we will also try and fine-tune some combinations on the day.”
However, the Thunder will be without their leader for Saturday's clash, with Owen unavailable for selection.
In other games, opening round winners Girgarre and Violet Town host Ardmona and Avenel respectively.
Murchison-Toolamba travels to take on Rushworth, while Merrigum welcomes rival Lancaster.
And Shepparton East and Longwood will fight for their first points of the season at Redlegs Stadium.
